Bereits in ihrer Kindheit in Neuschottland war die Musik eines ihrer Hobbies. Während sie an der Universität von New Brunswick Sport auf Lehramt studierte, nahm sie an einem Vorsingen für einen Spot in der wöchentlichen Fernsehshow "Singalong Jubilee" teil. Dort wurde sie jedoch abgelehnt, da bereits eine Altistin engagiert ist. Nach dieser Ablehnung legte Anne Murray ihren Hochschulabschluss ab und beginnt auf Highschool-Ebene Sport zu unterrichten. Zwei Jahre nach dem ursprünglichen "Singalong Jubilee"-Casting ruft der Produzent der Show, Bill Langstroth, sie eines Tages an und teilt ihr mit, er suche eine Altistin für die neue Show "Let's Go". Nach einiger Überzeugungsarbeit willigt Murray ein, ohne jedoch ihren Lehrberuf aufzugeben. Die nächsten vier Jahre singt sie für "Let's Go". In der Folge beginnt ihre berufliche Zusammenarbeit mit dem musikalischen Leiter der Show, Brian Ahern.
Murrays eigentliche Künstlerkarriere beginnt im Jahr 1968. Anfang des Jahres - sie arbeitet noch immer als Lehrerin - erhält sie einen Anruf von Ahern, der ihr vorschlägt, für das Independent-Label "Arc" Aufnahmen zu machen. Sie nimmt das Angebot an und spielt ihr erstes Album ein, das noch im gleichen Jahr unter dem Titel "What About Me" veröffentlicht wird. Das Album wird positiv aufgenommen und verkauft sich für ein Independent-Album durchaus gut "” so gut, dass Capitol Records auf Murray aufmerksam wird. 1969 unterschreibt sie schließlich bei der kanadischen Capitol einen langfristigen Plattenvertrag. Im folgenden Jahr wird ihre Debüt-Single für das Label veröffentlicht - "Snowbird". Die Single wird ein internationaler Hit und erreicht in Amerika die Top 10 sowohl in den Country- als auch in den Pop-Charts. Nach ihrem Erfolg mit "Snowbird" zieht Anne Murray nach Los Angeles, Kalifornien, wo sie nun regelmäßig in Glenn Campbells Fernsehshow auftritt. Der kalifornische Lebensstil behagt ihr jedoch nicht, und sie kehrt bald nach Kanada zurück.
1971 scheint es so, als ob "Snowbird" ihr einziger großer Hit bleiben würde - keine der folgenden Singles erhält große Aufmerksamkeit. Nur "A Stranger in My Place" knackt die Top 40. Mit einer Coverversion von Gordon Lightfoots "Cotton Jenny" kehrt sie in die Höhenzonen der Country-Charts zurück und erreicht immerhin Platz 11. Die nächste Single, "Danny's Song", steigt Anfang 1973 in die Top 10 ein, und zwar in beiden Kategorien, Pop und Country. Nach zwei kleineren Country-Hits kann sie Anfang 1974 mit "Love Song" erneut einen Top-10-Erfolg verbuchen. Auf die Single folgen zwei Top 10 Country-Hits - die Nummer-1-Singles "He Thinks I Still Care" und "Son of a Rotten Gambler". Nach diesen beiden Erfolgen kämpft Anne Murray einige Jahre lang weiter um den Einzug in die Top 40. In dieser Zeit widmet sie ihrer Familie größere Aufmerksamkeit als ihrer Künstlerkarriere. Sie heiratet Bill Langstroth und bekommt einen Sohn.
Die wirtschaftlich erfolgreichste Phase ihrer Karriere erfährt sie im Jahr 1978, als eine Coverversion von "Walk Right Back" bis auf Nummer 4 in den Country-Charts steigt. Kurz darauf folgt "You Need Me", ihr größter Hit seit "Songbird". Die Single steigt auf Platz 4 in den Country-Charts, klettert in den Pop-Charts auf Nummer 1 und erreicht Ende des Jahres Goldstatus. In den nächsten vier Jahren gelingt ihr eine fast ununterbrochene Serie von Top 10 Country-Hits, darunter die neun Nummer-1-Hits: "I Just Fall in Love Again" (1979), "Shadows in the Moonlight" (1979), "Broken Hearted Me" (1979), "Could I Have This Dance" (1980), "Blessed Are the Believers" (1981), "A Little Good News" (1983), "Just Another Woman in Love" (1984), "Nobody Loves Me Like You Do" (1984) und "Now and Forever (You and Me)" (1986). Die "Urban Cowboy"-Ära lässt Murrays Karriere prächtig gedeihen, zumal sie ihre musikalischen Einflüsse sowohl aus dem Pop- und Easy-Listening-Bereich als auch aus der Country-Musik bezieht.
Murrays Verkaufszahlen beginnen in der zweiten Hälfte der 80er-Jahre zu sinken. Grund dafür ist unter anderem der veränderte Hörgeschmack vieler Country-Fans, die nun zunehmend nach neuen, kantigeren neo-traditionalistischen Interpreten verlangen. Dennoch gelingt es ihr, sich mit gelegentlichen Aufnahmen und Konzerten in den späten 80ern und 90er-Jahren eine treue Hörerschaft zu erhalten ("Feed This Fire" wird im Sommer 1990 ein Überraschungshit und erreicht die Top 10).
2007 unterschrieb Anne Murray dann einen Plattenvertrag bei Manhattan Records. Das Label, das zur EMI Music gehört, veröffentlichte das von Kritikern hoch gelobte "Duets: Friends & Legends", mit Duetten von Martina McBride, Dusty Springfield, Nelly Furtado, Emmylou Harris, Celine Dion und vielen anderen mehr.
Anfang 2010 gab Anne Murray in einem Interview mit der kalifornischen Tageszeitung Contra Costa Times bekannt, dass sie zukünftig keine weiteren Alben aufnehmen werde.
